Unusual features in actinobacterial α-ketoacid dehydrogenase complexes: challenging old beliefs
Résumé
Pyruvate dehydrogenase (PDH) and α-ketoglutarate dehydrogenase (KDH) are conserved, tripartite enzyme complexes that carry out essential reactions in central metabolism. Despite their overall conservation, an increasing amount of evidence points to the presence of unique features in the structure and regulation of the actinobacterial versions of these enzymes. I will resume our ongoing studies on the components of a mixed PDH/KDH supercomplex from the species Mycobacterium smegmatis and Corynebacterium glutamicum, and show how our findings challenge old paradigms about the architecture and the regulation of these fascinating enzymatic machines.
